Luciferase assay - (Jul/08/2009 )

Hi, i like to know if anyone has a suitable recipe for lysis buffer that could be used to lyse cells for luciferase assay not those from commercial kits. Also anyone has recipe for the stop solution for luciferase assay. Thanks in advance

Candice,

A common buffer for cell lysis prior to Luciferase is CCLR buffer, which is 100 mM potassium phosphate, pH 7.8, 1 mM EDTA, 7 mM 2-mercaptoethanol, 1% (v/v) Triton X-100, and 10% (v/v) glycerol.

I don't know the recipe for stop solution, sorry.

you can use normal np-40 lysis buffer

have a glance at
