Beta Catenin immunofluorescent staining - (Jun/23/2009 )

Hi all,

I'm trying to stain for beta catenin to visualize it's location (cytoplasmic vs nuclear) after treatment. But I keep getting punctate stains...

I am wondering if it's my fixation protocol, I used 10mins in 4% PFA.
I'm thinking of switching to cold-Methanol.

If anyone has experience with this, I would really appreciate some advice. Thanks!

I have used pre-chilled 1:1 Methanol:Acetone fixation for 5 min at -20 and 10 min room temperature, 0.2% Triton-X treatment for mammary epithelial cells for B-Catenin staining which worked fine.
