neuronal nitric oxide synthase/cell line - (Jun/19/2009 )

Hi folks,

I am searching for a human cell line that expresses high amounts of neuronal nitric oxide synthase. Has anyone worked with such cells and can give me a hint.

Is there a reason that you can't take a neuronal line and make it overexpress nNOS? Just an idea...

Yes


High levels of NO are cytotoxic to most cells.......neuronal cells/lines are in general more prone to damage than other cells such as Macrophages, endothelial/epithelial, smooth muscle cells

you can titer expression by just transfecting less or even infecting at an MOI of 1 or less. if neurons can't take this, chances are you won't find neurons that express nNOS at that level.
you can also use an inducible system to turn on or off nNOS expression.

.. well unless there is something else that causes a problem with nNOS transgene expression.
