RNA contamination in PCR - (May/26/2009 )

Hi all,

I am trying to amplify microsatellite markers from genomic DNA, however I constantly fail to get a band from the PCR.

I did not perform RNase treatment on the DNA prior to PCR. Will RNA contamination be the problem? I've searched online on how RNA might inhibit or hamper PCR reactions, but could not find anything convincing.

Please help!

We do PCR on concurrently extracted DNA and RNA from the same specimens all the time. You shouldn't have any problem with RNA in the DNA sample. You might want to do troubleshooting on other PCR parameters, at least do a positive control to ensure your PCR works.
