TOP10F' - Can they be used for regular vector propagation? (May/21/2009 )

We've got several boxes of those cells accidentally bought by one of the undergrads and I'm not sure if they can be used for regular vector propagation. I would assume that if the construct of interest doesn't have the f' origin, it should be fine to use them as regular competent cells. I wonder what would happen if vector has this f' origin. I've never worked on ssDNA production, hence lack of knowledge. Do you think that normal replication would take place? Do you think that yield could be decreased because of that? Any thoughts are welcome.

For normal cloning, there should be no problem. If you work with BACs, then the BAC origin is also an F origin, and you may have a problem.
