culture C6/36 insect cell lines - (Apr/30/2009 )

Dear friends
I would like culture C6/36 insect cell lines. But the recommended temperature for culturing this cell lines is 28 oC with 5%CO2. we have incubator in our lab. with 37 oC but without 5%CO2. Has anyone advice my to culture this cell lines.

I assume you are planning to grow them in media like RPMI 1640 or DMEM that require 5% Co2 for buffering. The line is easily adapted to growth in more classical insect medium like Graces +10%FCS or Sf900II (Invitrogen). The advantage is that these media are designed for incubation without CO2.
As for temperature, most insect lines grow reasonably well when incubated at "lab temperature". If you cannot find an old fashioned oven set at 27 oC, try incubating them (in the dark) on a lab shelf. For most purposes they should be fine.
