selection of internal control gene - (Apr/29/2009 )

I am going to do qPCR. My target gene is expressed at a very low level. Can I use housekeeping genes? I was told that comparable CTs between target and control are preferred, but can't find any official saying about this.

It is more important that your amplification efficiencies for both genes (target and control) are similar, rather than their absolute Ct values. Some of the housekeeping genes should be fine.

I would suggest using more than 1 house keeping gene just so that the results donot mislead you.
