plaque assay haven't worked - (Apr/09/2009 )

hi Friends!!!

Everybody knows that Vero cells are commonly used in virology methods. I have tried for many times to make plaque assay with Vero cells but actually they are not working!!! in the past they did. i have other cells lines like LLC-MK2 and Hela and when the plaque assay are performed with these cells they are greatly good.
My question: why vero cells have not been working in plaque assay?

I'm very surprised to hear this, because I couldn't get plaque with HeLa so I decided to switch to Vero. Vero must work better I guess but it depends on the virus too. In the end I figure out not to have very confluent wells.

Is your virus a human virus or avian or..?

Wow that's surprised too!! I work with Flavivirus, specially Dengue Virus and Yellow fever Virus. They frequently hit 105 PFU/ml
Let me say:Is it 160 passage, Vero got old?
