One protein, known function, known motifs. If isoelectric point of one of the motifs is changed, eg. 6 to 8, will that affect protein function?

"Maybe" is the best answer I can give you. You'll not know until you try it.

I've never heard of applying an Ip to a motif; I've only used Ip's in the context of the whole protein.

If a change in function occurs, it will be due to the change in amino acid sequence, which will also cause a change in the Ip. You can't necessarily attribute the change in function, if any, directly to the Ip change.
