Hi all, i need help?

my study on dengue virus, i want infect monocytes by dengue type2 by use MOI.

in published article was used MOI of 0.1

what is the volume of virus use into infect monocytes?

what is a protocol use to determine MOI?

How calculate MOI?

MOI is __m__ultiplicity __o__f __i__nfection

which essentially means the number of infectious particles (ie. your virus) per cell

eg an MOI of 1 means that there is one virion for every cell, MOI of 0.1 means there is one virion for every 10 cells.

So you need to know what the titre of your virus is, and how many cells you have- then its pretty simple to work out how much of your virus you should use.

Its the same.

Its just how many bacteria per cell..

And what do you mean by calculating? Normally its something you pick yourself.

(you decide how many bacteria you will use to infect a certain cell or culture)

I am asked to add my bacteria at an OD of 0.01 (exponential phase). I am wondering if my MOI can be calculated from this.

I do not understand what you mean?

You can not link OD with MOI unless you mean that your MOI is the OD 0.01 and you know how much bacteria there are in OD 0.O1

BTW: OD 0.01 ? Thats nothing...
