Monocyte - (Mar/23/2009 )
Hi all,
I have a questions about monocyte unfortunately nobody couldn't answer them, i hope if anyone could help me.
If we have a look on the THP-1 and AML-193 cell line (Both are acute monocytic leukemia and the cell type is monocyte) we find:-
THP-1:- split 1:2 to 1:3 every 3-4 days and the doubling time around 35-50 hours.
AML-193:- split 1:2 every 2-3 days and the doubling time around 50-60 hours.
I have read that monocyte is non dividing cell so why is it dividing in the two cell line?
Is it dividing because it's leukemic cell?
If i separate monocytes from acute moncytic leukemic patient then culture them, can i split them?
If i separate monocytes from healthy donor then culture them, can i split them too?
Any help is highly appreciated

hi,
i thank mature monocytes from peripheral blood can not proliferation bzc not cell slf-renweal like stem cells.
in this case i thank should be isolation stem cell and proliferation, and fial differentiate into monocytes by use cytokine IL 3 and growth factor GM-CSF

I got the answers for my questions.
Since acute monocytic leukemia patient has monoblasts and promonocytes, i'll isolate the cells using CD14 Microbeads because both cells express CD11b, CD11c, CD13, CD14, CD45bright, and CD64 then split and differentiate.
