what is meant by passages - (Mar/17/2009 )

Hi

i wanted to know exactly what is meant by passage in cell culture and how many passages can be max. done for Nalm6 cells ? :(

The term passage means to split the cells down so they dont become overpopulated. In my experience this can be done 9 or 10 times before the cells begin to become useless but for your specific strain I dont know :) I used COS-7 and another more delicate type of cell (the name escapes me!)

Unless you are using primary cells or have a very sensitive endpoint, 9 or 10 passages shouldn't be the limit. For my system, various dose response parameters (like Vmax and EC50) are consistent oftentimes beyond 200 passages for COS-7, CV-1, and U2OS cells.

I like to just keep my cells around for 2 months or 24 passages, whichever should come first.
