lysis buffer to use for enzyme assay - (Feb/22/2009 )

hello,

i'd like to do an enzyme activity assay comparing treated/untreated cells. is there a particular buffer i should use to lyse the cells but not denature the proteins?

Yeah

I use 50 mM Tris pH8 with 10 % glycerol or 100 mM phosphate pH 7.2 with 10 % glycerol. Also, u may have to add other things like ATP, CoASH or NAD(P)H if yr enzyme needs it.
