passage number for cell line - (Feb/11/2009 )

Hi,
I am wondering how important is the passage number cell lines?

As far as i know each cell type has a particular range of passage efficiency if it crosses beyond that cells behave abberantly. I faced problems like poor transfection efficiency and decreased doubling time.

Keep track of passage numbers and after a certain number of passages, they change many of their properties at which point, you throw them and get new ones out. Each cell line is different with regard to passage number.

passage number is very important.

After a while, the cells begin to behave differently and may not give you consistent results.

If you get cells from ATCC, they might have something mentioned about the passage number in their product information sheets.

I had noticed morphology and growth rate change in late passage cell line.

my boss just asked for a cell line from other lab and let us grow it. I am wondering how old this cell line is and maybe we should buy some from ATCC instead.
For 293 cells, how many times do you guys usually pass them?

I have had different batches of 293 and they all behave differently. Usually I see the difference after transfection and thats when I decide to get new cells. Normally I have the 293's for 15-20 passages.

Better to have them from ATCC than other lab. Also test cells for mycoplasma when you get from any source.

That's what we did in my previous lab, I think I should talk my new boss into buying a new patch of 293 cells from ATCC.
