galactose induction of yeast cells - (Feb/01/2009 )

Hi All.
I am trying to express my genes in yeast dual expression vector. I got colonies after transformation of yeast with the vector in glucose containing medium. But when I transferred the cells to galactose containing medium for expression of my genes under GAL promoters there was no growth. Control (vector only)is not growing too. Can anyone provide me the protocol for galactose induction.
Plzzzzzzz help me....

Amita

Yeast take longer to grow in galactose media than glucose, have you left them to grow for 48 hours?

Yeah!!!
I have grown them from 16-72 hrs but no growth was observed.
How to grow then, even control is not growing.
