Standard HCL protein hydrolysis technique without vacuum - (Mar/31/2020 )

Hello everyone,

 

I am trying to follow the standard protocol for digesting proteins to amino acids. The hydrolysis needs to be done under vacuum. As I don't have an access to the vacuum setup, can I just purge my tubes with nitrogen and incubate them as recommended for 24 h at 110C?

According to the procedure to which you link, acid hydrolysis can be performed by reflux or in a sealed tube either in vacuum or nitrogen. It’s been a long time since i performed the procedure and don’t remember but i think i did it under vacuum (i only did it 2 or 3 times).
