LiAc transformation - HELP! - (Oct/26/2018 )

Hi guys, 

 

if anybody has a copy of LiAc transofrmation' protocol from Methods in Yeast Genetics: A Laboratory Course Manual, Cold Spring Harbor NY (1994, Kaiser et, al.).

 

 

I have a problem with transformation - I received a plasmid  which overproduce SLA1. I want to transform strains which have deleted genes involved in assembly in the cytoskeleton. In the paper is written that they used transformation protocol from that book but unfortunately I can't find it anywhere .

 

I tried many protocols but none work. I really want to test protocol from that book. 

 

Please, help me! You are my last hope.

Thank you for your help, I'm very grateful.

I used this method and didn't work with my plasmid. I realy want to find method from Methods in Yeast Genetics: A Laboratory Course Manual, Cold Spring Harbor NY (1994, Kaiser et, al.) becouse I want know what they did diffrent to achieve the success.

This is a job for your friendly neighborhood Librarian! They can often get access to things the rest of can't, through Interlibrary Loan.  I'd recommend a visit to your University library!
