Crosslinking with glutaraldehyde - (Jul/24/2017 )

hi,

 

Im trying to crosslink lipid with amine group and a modified sugar with amine group. When I add Glutaraldehyde to the solution of lipid and sugar I see a colour change to yellow. But on analysis with tnbs Im not getting any result. please help

Dual NHS linkers would work for this. Thermo offers a variety of these: https://www.thermofisher.com/us/en/home/life-science/protein-biology/protein-labeling-crosslinking/protein-crosslinking/amine-amine-crosslinkers.html

 

Note you will get homo crosslinking as well as heterocrosslinking with this chemistry.

 

Use orthogonal functional groups instead of amines to prevent this, such as azide on lipid + alkyne on sugar (click chemistry) or TCO + tetrazine, etc. Either synthesize these directly to you lipid and sugar, or If you don't mind long linkers, you could use what you have and add NHS--azide linker to sugar, NHS-DBCO linker to sugar, then crosslink lipid-azide to sugar-DBCO. Cruise through the Thermo website or "Click Chemistry Tools" to see what options are commercially available.
