Crosslinking with glutaraldehyde - (Jul/24/2017 )
hi,
Im trying to crosslink lipid with amine group and a modified sugar with amine group. When I add Glutaraldehyde to the solution of lipid and sugar I see a colour change to yellow. But on analysis with tnbs Im not getting any result. please help

Note you will get homo crosslinking as well as heterocrosslinking with this chemistry.
Use orthogonal functional groups instead of amines to prevent this, such as azide on lipid + alkyne on sugar (click chemistry) or TCO + tetrazine, etc. Either synthesize these directly to you lipid and sugar, or If you don't mind long linkers, you could use what you have and add NHS--azide linker to sugar, NHS-DBCO linker to sugar, then crosslink lipid-azide to sugar-DBCO. Cruise through the Thermo website or "Click Chemistry Tools" to see what options are commercially available.
