GFP tagged plasmid - (Oct/12/2016 )

HI,

I ordered a GFP-tagged plasmid from Origene recently for stable cell line establishment. However, after 24-48 hr transfection I cannot observe green signal under fluorescence microscope. I have sequenced the plasmid using the origene supplied primers and there is a GFP tag sequence.

Has anyone experienced something similar? Any advice would be appreciated.

There could be any number of problems. Can you give us details of your experiments, such as how much DNA was transfected into how many cells in what vessel? Did you check the GFP for an ATG etc.?
