Primer sequences with M and Y - how to order? - (Feb/17/2016 )

Hi,

    I have some primer sequences which I have to order. Got this from a paper, and these sequences have 'M' and 'Y' (mixed nucleotides) at some positions. How should I order? as it is with the M and Y or replace them appropriate nucleotides? Plz advice... Regards... Ratnam.

This probably depends on where you order from, but IDT, for example, will gladly synthesize with the M and Y in the sequence. You should be aware that the ratio of the bases in those positions are likely not exactly 50% due to inaccurate mixing of the reagents and differences in their reactivity during synthesis.
