CHO-K1 media - (Dec/02/2015 )

Hello, I require some help.

I'm trying to make media for CHO-K1 cell line. I have DMEM without amino acids or FBS.

First I need to make 50 mL of 100X L-Proline solution stock for future usage. However I don't know how to approach this calculation. I have powder L-Proline, and the formulation for the media is to contain L-Proline 0.06906 g/L.

Here is what I did:

(0.06906 g/L) -> (3.453x10^-4 g/0.05L) x 100 = 0.03453 g/0.05L for a 50mL of 100x L-Proline stock solution. Is that right?

The easiest way to do calculations is to be transparent at all steps... use formulae to show working, just like you were shown in school!

 

The formula you want here is c=n/v rearranged to get n=c*v

 

n= 0.06906 g/l * 0.05 l

n=0.003453 g

 

multiply by 100 to get 100x solution

0.003453 * 100

=0.3453g

 

Note that both the ATCC and ECACC recommend Hams F12 for CHO-K1. Check the appropriate medium with the people you got the cell line from, as they may have weaned it onto DMEM or not!
