Stock solution: preparation from chemicals? - (Nov/14/2015 )

"A basal inorganic medium (pH 8.0–8.2) for enrichment and isolation of ammonium oxidation bacteria (AOB) was prepared by dissolving 0.5 g ammonium sulfate, <(NH4)2SO4 as a source of reduced N to investigate the acceptability of inorganic N sources for heterotrophic nitrification;  0.7 g KH2PO4, 0.5 g MgSO47H2O, 0.5 g CaCl22H2O and 1 mL of trace mineral solution in 1 L of distilled water. The trace mineral solution contained (per liter): 2.86 g H3BO3, 0.22 g ZnSO47H2O, 0.08 g CuSO4 5H2O, 2.03 g MnSO44H2O, and 1.26 g Na2MoO42H2O (pH 7.0). The AOB medium was autoclaved for 30 min at 121 C."

Help, how do I prepare the stock solution from the chemicals? Do I mix them all in one step or should I have a way of mixing them? I'm having difficulties finding protocols for stock solution creation for ammonia-oxidizing bacteria medium, as all that they show are recipes and not how i should create them. thank you so much

First make the stock trace mineral solution. You can make only 100 ml (or less) since you probably don't need a liter. Divide all the weights by 10, add them to a bottle, fill with pure water to 100 ml and dissolve. You may have trouble weighing the copper sulfate and may need to make a more concentrated solution of just that and add it, or just add a few crystals and call it good.

 

Then, add everything else and 1 ml of your trace metal solution to a bottle, bring to 1 liter with pure water, then autoclave it. Sometimes there are issues with salts not dissolving or forming precipitates, but none of these sound like they will do that. Problem components tend to be iron compounds and precipitating phosphates.
