yeast two hybrid 3-AT - (Oct/31/2015 )

Hey all,

I was reading about the use of  3-AT in the yeast two hybrid system. They use it to lower false positive as 3-AT is a competitive inhibitor of the his3 gene product.

But what if you would use another product that is a competitive inhibitor of another protein in the histidine pathway?
Or the other way around: if you use another reporter gene in that pathway and still use the 3-AT ?

Would this still be possible or?

I saw a paper where they used another reporter gene (his1) and they did not use 3-AT, which made me wonder about it

I think using another inhibitor with the same reporter would work.

But a different reported with 3-AT wouldn't work, because 3-AT is specific to his1.

 

What are you thinking of specifically?
