Confused with the CALCULATION statement - (Sep/06/2015 )

Hello,

I am getting confused with the Calculation statement given in the ELISA protocol.

This is for preparing reagents and it says that:

"Dilute 50 ml of Wash Buffer Concentrate into de-ionized or distilled water (dH2O) to a final volume of 500ml". Does this mean I take 50 ml of wash buffer + 450 mL of water

OR

Does this mean I do 50*x = 1*500

x = 500/50 = 10 ml of wash buffer + 490 ml of water

Kindly advice.

It means your first option.  Frankly, I can't imagine how you think it could mean the second.
