Chemical to revert actin aggregates - (Jun/27/2015 )

Hi all.

I'm looking for a compound that can revert the F-actin aggregation defect that I observe in my cells. Any thoughts?

you can depolymerize f-actin outside of the cell with atp and magnesium in a solution that has no sodium or potassium chloride added. this low salt condition may cause problems with cells.

 

what f-actin defect? actin should be filamentous in the cell.

I observe the formation of large aggregates of filamentous actin (chunks of actin) instead of cables.

 

My hypothesis is that this defect might be causing other downstream phenotypes. So my idea would be to treat the cells with a compound that could revert the actin aggregates and that cells would recover from those downstream phenotypes.

 

Maybe with a low dose of a depolymerizing agent like latrunculin-A?

under what conditions do you observe the aggregated filaments?

 

latrunculin may have too much effect.
