Jurkat cell - normal receptor/gene expressivity - (Jun/21/2015 )

Hi,

 

I'm new to using Jurkat cells and wanted to ask whether transfection was necessary to study non-IL-2 characteristics of Jurkat cells. For example, can they express FOXP3? CTLA-4 or PD-1? or would I need to transfect these genes into the Jurkat cells prior to study?

 

Also, I've plated my cells out and was wondering if I need to continually split them or if I can just replace the media directly? I'm just worried since the cells are in suspension I'll aspirate the bulk of the cell when replacing the media.

I would do a Flow cytometry analysis on the cells to check for the expression of those targets.

 

Yes, you should be splitting them by taking a fraction of the cells from your culture and adding them to fresh media.
