Antisense vectors confused about construction - (May/27/2015 )

When constructing antisense vectors for gene silencing, I'm a bit confused about the antisense sequence.

Do I just need to reverse complement this as if I was making a reverse primer?

Or am I just supposed to complement it?

What would happen in either case - which one binds with the mRNA produced from the sense sequence?

It is essentially NEVER correct to use the complement nstead of the reverse complement. Pretend you are designing the reverse primer.
