Does TritonX-100 lysis buffer break up mitochondrial membrane? - (May/09/2015 )

TritonX-100 Lysis buffer recipe = 25 mM Tris•HCl pH 7.4, 150 mM NaCl, 1% Triton X-100, 1 mM EDTA, 5% glycerol

 

I am sure it won't break up nuclear membrane but not sure whether it can break up mitochondria membrane.

yes it does (personal experience)

sodium deoxycholate is used for nuclear membrane.

Thank Curtis. I am confused about this for some time, because some of the protocols (eg. ABCAM) states that mild lysis buffer like tritonX-100 won't lyse/break mitochondrial membrane. SDS seems to be the one to break mitochondria in RIPA buffer.
