What is best to measure protein expression: western blot or ELISA? - (Apr/16/2015 )

Hey guys,

I have a question. I want to measure the expression levels of some proteins. Here in my lab, people usually do western, but it is not good as a quantitative method and is also a little expensive (if you buy the reagents). 

I never worked with ELISA, but this method may be a good choice to evaluate the expression levels of some proteins in my project, it can evaluate a lot of samples at the same time, is cheaper and seems to be quantitative. Is it true? Does anyone with experience with elisa and western can help me to clarify these questions?

elisa is a quantitative method. western blot is more qualitative than quantitative (it can be quantitative within limits).

You might also consider your samples; are they pure protein-of-interest, soluble or not, and some other factors you may put in mind when choosing the suitable method. 

ELISA is good way to quantitate protein in different matrix, such as serum, plasma, cell culture supernatant, tissue lysate. Sometimes, Antibodies work well in western blots, but they may not work in ELISA.  In terms of time and effort, normal ELISA is comparable to western blots. Now, one fast ELISA ,called EasyTest ELISA , just needs 1.5h to finish the whole process, significantly saving time and effort. So, you may finish your large amount of samples within one day.
