Unbroken cell - (Mar/27/2015 )

I have worked on bacterial cell can resist breakdown even by french press or sonication. Have anyone suggest rush technique for such cell?

No I haven't found a rush technique but  I use a pretreatment that have always functioning, if your bacteria is a gram +  the pretreat buffer:20mM tris cl ph 8.0, 2mM sodium efta, 1.2% triton x-100 you can prepare this buffer ahead and just before use add the lysozyme 20mg/mL

resuspend pellet 180microL of the pretreatment buffer and incubate at least 30 min at 37C.
