how to confirm lentiviral integration in a target cell line - (Mar/24/2015 )

i am using the plko system,and  how can i  confirm lentiviral integration in a target cell line?

1. Cells will survive puro selection which means the virus integrated. 

2. You can design primers that target the integrated virus (does not matter what you target). Then run PCR on the isolated genomic DNA from your cells. 

3. You can also determine where the virus integrated. First, you would need transduce your cells making sure only one viral integrate per cell then establish isogenic clones.
