RNA extraction from oocytes - (Mar/20/2015 )

Hi, everyone. I'm a student who is doing my final year project in my degree level. I need to extract the RNA from oocytes from mouse. However, there is seems no band when I run electrophoresis neither for my cDNA product nor PCR product. Is there any workable protocol suggested? Thank you for your help. =)

I'm using TRIZOL reagent method to extracted RNA. Is it possible if I do not add in glycogen to extract RNA?

How much RNA you will get and whether you should use a carrier such as glycogen depends on how much input you are using.  If you haven't done RNA extraction before, I would recommend trying it on other tissues to see how it works, and to ensure that you are capable of getting RNA out the end before you try it on limited samples (size, not availability), such as oocytes.

 

If you are trying to extract from only a few oocytes, you are unlikely to get enough RNA to visualize on a gel, and as such you won't see anything on the cDNA prep either. Make sure you can get the PCR working before you try it on your oocyte cDNA - it is likely that this is failing, not because of the input cDNA, but for some other reason.
