Please help - (Mar/09/2015 )

Hii... i am culturing THP1 cells in RPMI 1640 (as advised by ATCC formulation, except 2ME). Unfortunately my cells are forming clumps few hours after seeding (at 1* 10^5 cells/ml) before even going to 60% of confluence. I am using pen-strep in the media to prevent bacterial contamination. After they form clumps, they used to stop growing. How can i get over this problem? plz help!!

does your cell medium contains serum?

Yes I use 10% heat inactivated (one hour at 55 degree C) FBS from authentic source...

Dear Mr. Das, Why not try shaking theculture every few hours (~ 1 hour/3 hours). Lueukemia cells after all are not sedentary cells. Thanks.
