storing membrane - (Dec/22/2014 )

Hi,

 

Our dark room to develop the western blot film is out of service for 4 days.  I have finished washing my membrane after the secondary antibody incubation. Will it be ok to store the membrane in 4deg and deteft the signal after 4 days?  Has anyone done  this?

you should be able to store the membrane wet with blocking buffer or pbs or tbs.

 

if you know how long you need to expose then you could expose the film, remove it from the blot and store it in a different cassette until you can process.

Depends on your signal. Some signals are so strong they can burn an image in the membrane. But I concur with mdfenko; just expose several films at different intervals as you would usually expect and store the films in dark until such time as you can process them.

 

Sure I know your machine has been working for last 20 days, but then I like dispensing advice left and right, especially when I am fighting a crave for a heavy heavy lunch to destroy my new years resolution.
