Detection of Td-Tomato protein in frozen sections - (Dec/09/2014 )

Hello,

We are working with  Wnt1-cre:Rosa^{tomato mice} and have trouble detecting red fluorescence in frozen sections. Any explanation, any tips?

What processes are you doing to the sections before viewing?

fixation before freezing kills most, if not all, fluorescence of GFP etc, interfering with the resonance of the beta barrel. I'd recommend antibody staining against tdT, there are some from Clontech for example.
