Protect agar plates from 55C temperatures - (Dec/05/2014 )
Hello
I am a bit concerned.
I tried grow Bacillus but the plates quickly desiccate and agar shatters. Anyone has any tips on what I could do to protect the plates from quick desiccation? Anyway I had growth so if the plates stayed there a bit more I would have a proper culture of Bacillus
55 is pretty high. But you can put the plates into a plastic bag, which reduces evaporation, or seal them with parafilm. You can also humidify your incubator with a tray of water.
Yes,
You can do what Phage434 told you.
Myself: I pour the plates thick enoug, wrap parafilm around it and that is normally enough.
If I have to grown them for a real long time, I just put some water in the incubator...
great!
I need to grow some Thermus and Bacillus sterothermophilus. Needs 65 and 55 degrees so... these tips are going to be helpfull
Thank you!
this is for real extreem cases:
I put my plate in a different (bigger) plate that is filled with water (make sure your plate with cells itself stays "dry", I mean: that the water does not enter your plate) and put my plate is this bigger plate that you can seal with a lid..
Not sure if you get what I mean? Its a plate in a plate...
And if you use sterile water and prepare this in the flow itself, its a closed sterile environment...
