SaOS-2 Minerlization response. - (Dec/01/2014 )

Hi,

 

I've been using SaOS-2 cell line for determining the effect of my drug on its mineralization ability. The problem I'm currently facing is that the cell line which previously showed low minerlization activity at a particular concentration of my drug is now showing mineralization activity similar to that of the positive control in the presence of my drug. How to fix the inconsistent result I'm getting. Any help will be deeply appreciated.

Did you change the batch of drug? Checked expiry, storage conditions etc? I don't know how experienced you are at cell culture, but cells need to be handled in a very consistent manner (numbers, media, pre-treatment culture conditions etc).

 

Importantly, SaOS-2 cell line, with more passages increase in its mineralization activity (here). So if you are going at it for months, things could change. Freeze aliquots at regular intervals to avoid late-passage cells.
