Ligation and blue white screening - (Nov/28/2014 )

Greetings

 

Im working with two pTZ57/R plasmids (A and B). Each plasmid had a fragment inserted into its MCS therefore the beta-galactosidase gene has been interrupted. next I then cut a portion of each pTZ plasmid, this portions contain the insert and part of the pTZ sequence, i then ligate the two portions together. Now, after transformation, do i plate onto IPTG XGAL plates or just normal plates, becasue i mean the beta galactosidase gene has already been interrupted so there wont be any blue colonies formed right? so i can just go ahead and plate on ordinary amp plates?

Without seeing the sequence it is hard to tell, but it sounds like you can just plate on regular plates.
