Tissue homogenization using Potassium phosphate buffer - (Nov/17/2014 )

Hi Friends,

 

I want to know one thing that, mostly we will be using Potassium phosphate buffer for homogenization to measure enzyme activity in tissue.

My question is whether this homogenization procedure ruptures all the organelles like mitochondria, Golgi etc.

Can we use this protocol to measure activity of mitochondrial enzymes?

that depends on the constitution of the lysis buffer and the homogenization conditions (physical as well as chemical).
