protein G - (Oct/13/2014 )

Hi I obtained Antibody purification kit(protein G) and developed my lysed serum (mouse serum is red due to have hemoglobin ) but my cartridge do not passed my sample and we washed and eluted our sample in upper of cartridge. of course, we used only 100µl of resin . we think the hemoglobin is problem.

1-plz help me about treatment way of serum method ?  

2-could we reuse this cartridge or If not , may i used another cartridge ,whatever.

the hemoglobin should not affect binding of the antibody to protein g, you may have to wash more to remove all of it before you elute the antibody.

protein g should be reusable as long as you don't use overly harsh conditions to elute the bound antibody.

are you sure you need protein g and not protein a to bind your antibody?

have you determined that the antibody bound to the protein g and didn't just elute in the wash?
