Inhibitors for kinase inhibition experiments - (Aug/11/2014 )

Hi all, there are inhibitors for many kinases like Akt, CAMKii

I recently wanted to inhibit these. My questions are following:

 

1. what does it mean that they used 10 microM of inhibitor KN93 (in literature)? Did they use 10 microM inhibitor per litre?  this means, if i am trying to inhibit it in 10 ml medium in 10 cm cell culture plate, i am supposed to add 1 microM per 100 ml medium, which would be almost 1/10 microM per 10 ml medium? ohmy.png!   Am i right or wrong?

 

2. Do you guys serum starve cells before u add inhibitors? if yes, how long and after serum starvation, do u remove serumfree medium and add serumplus medium and then add inhibitors  or you add inhibitors in the same serum-free medium which the cells were in.

if it is written uM (capital M) then it is usually considered to be a concentration (i.e. umol per litre).
