Differentiation of human cortical neurons - (May/22/2014 )

hi

I want to grow and differentiate human cortical neurons. I have a vial of HCN-2 cells from ATCC. From what I understand, they are extremely slow and difficult to differentiate. Changing my cell type is not a viable choice right now. So I make do and hav a couple of questions. If anybody has any experience either differentiating these or other cortical neurons, I would really appreciate some help.

* The nerve growth factor required to differentiate these cells do they have to be human or mouse would be ok as well? ( apologise if this is a really dumb question!)

* ATCC is no help answering anything as they were stored there and ATCC never differentiated them.

* Do you add the cAMP, NGF and IBMX required for differentiation in the media or fresh to the cells every time?

* is there any coating of Matrigel or collagen I required to help the cells grow better?

Electrophysiological whole-cell voltage- and current-clamp recordings were performed on two control and three FXS patient lines of human cortical neurons derived from induced pluripotent stem cells. In addition, we also describe the properties of an isogenic pair of lines in one of which FMR1 gene expression has been silenced.
