Do you go by 'total protein' or 'tissue weight'? - (Apr/09/2014 )

Hi All, 

 

I am using a sandwich ELISA to measure protein from homogenized tissue. I have a couple of basic questions:

 

1) Do you report your ELISA results 'per tissue weight' or 'per mg protein'?

2) Do you load your wells so as they have equal protein, or so they have equal tissue weight? 

 

I have been told that loading either way is fine, but am wondering, in general, if you have a preference and why?

Either is fine - they are both equivalent - the percentage  protein you extract from one sample should be same as for another, assuming you are sampling the same tissue type each time.
