Conjugation of Ab to magnetic beads to isolate T cells from Spleen - (Mar/04/2014 )

Hi there,

I want to isolate CD8 T cells from spleenocytes, I have both magnetic beads and a normal anti CD 8 antibody with me. Is there a way I can conjugate this Ab to the beads to get a pure population. After conjugation can I follow the normal procedures laid down by MACS for seprating cells?

Thanks in advance.

are the magnetic beads functionalized?

Your magnetic beads need to be either carboxylated or precoated with avidin (which requires a biotinylated antibody).

Here is a guide for coupling protein to microspheres / beads:

Luminex provides magnetic beads that are funtionalized.
