I always have a cell depris floating in the supernatent of MDCK cells - (Feb/26/2014 )

Dear All, 

 

I am growing MDCK for titrating Influenza virus. Really tricky, I always have after one day a floating cell depris in the supernatent of cells. I used 10 % FCS, 1 % pyrivate, 1 % Glutamate in MEM medium. What might be the cause ? 

 

Is there is other cell types that I can make plague assay for influenza virus other than MDCK cells ?

Vero is the best one. Also you probably culture too many cells that some don't find room to grow, or they are already dead when you culture. One reason could be that you scraped your cells?

Could you supply me with a protocol to titrate influenza with VERO cell. I propagated the virus in MDCK cell, Is it true that I can titrate it in another typr (VERO)

plaque assay can be used for many viruses. The protocol is available on internet. most of them are similar. here I don't have one to give you. I used vero for NDV infection. but bare in mind not all cells are suitable for plaque assay. Never use HeLa.
