Live cell sort DNA stain permeabilization - (Jan/29/2014 )

Hi, All,

I am planning using Chromomycin A3 for live cell sort based on DNA content because that is the only dye I've found so far stains specifically for DNA, and does not require UV wavelength (<400nm) for excitation. But I am not sure what is a better choice for permeabilizing the cell/nuclear membranes? Triton X, tween 20, or digitonin? What would be a good concentration ? I am using HeLa cells. Anyone had any experiences?

As far as I am aware you would need triton to puncture the nuclear membrane.  However, I think this would also irreversibly damage both the nuclear and cytoplasmic membranes.
