What method is the best to measure Telomeres? - (Jan/16/2014 )

Hi everyone !!

I just find a correlation about my protein and telomeres length =)

So my next step is increase the number of sample. I have about 200 sample from patient.

I have read about qPCR and Flow Fish.

somebody know what is more cheap?

I understand for qPCR i need primers and no more (Syber and termocycler of course)

and more easy-quick?

somebody use it?

Can i use Flow Fish in quiescent cell? Can i use Flow Fish to tissue?

if I wanna compare cell culture too. What is the best method?

I have experience in qPCR and i understand very well the protocol by Cawthon but i am beginner in FACS.

I don't know if this is outdated technology or not, but a colleague of mine used to use TeloTAGGG kits from Roche.

these kit provide 50 reaction for 770 €

i have 200 patients...mmmm to expensive =)

Golden standards for measuring telomere length are TRF analysis, which includes subtelomeric sequence as there are no restriction sites in TTAGGG sequences or telomere FISH with PNA probes via hybrifisation either in situ on fixed cells on a slide or in suspension and analyses by FACS analysis (Flow-FISH).
