seeking for FISH handbook etc - (Jan/13/2014 )

Can someone redirect me please for a good free resource for a FISH handbook or even a website where i can find

 

the basic of FISH and the principle behind it. for example - when you pre-wash slides with 2Xssc, what does it do to a cells, tissue etc?

You might also want to visit Tavi's  FISH Guide.  I have found it quite useful most of the times.

thx for all answers, but still i'm looking for some even more detailed info about the FISH technique. Tavi's  FISH Guide is great but not detailed enough for me.

 

any how, can someone answer me if 70% denatured ethanol damage FISH on cell suspension?

I'm doing FISH on cell suspension from sputum and would like to know if ethanol denatured can damage Hybridisation (weak signals) or not?

when you are washing your sample after application of the probe and the hybridisation duration, ethanol is merely helping you dehydrate the slide. It does not have any effect on the signal strength that you are getting. For weaker signals, your should look at the sample preparation, denaturation and hybridisation steps for slip ups.
