Can multiple genes follow only one promoter? - (Dec/26/2013 )

I'm planning to express 4 genes (total length ~9 kb) under one promoter in Clostrdium. Each gene will have its own "aggagg" RBS. Is this feasible? Is it gonna influence the strength of the expression? Is there anything special I need to pay attention to? Thank you for your help!

Yes, this is feasible. As long as you have no terminator sequence within or between the genes, transcription will proceed. In prokaryotes, it is common to see multiple gene operons. There may be substantial variation in the transcription levels and protein expression levels of the different genes, so if this is important, you may need to optimize the relative RBS strengths.
