Genomic DNA and plasmid DNA isolation - (Jun/16/2013 )

HI, i will be doing whole genome sequence of a bacteria. And for that i need gDNA and plasmid both in the same isolate. I am using Promega DNA purification kit for DNA extraction. Is this kit able to isolate bothe gDNA and Plasmid in a good amount. Is there any other, method for isolation. Please suggest.

The easiest gDNA (and plasmid) extraction I'm aware of is the Zymo kit, which is also available in 96 well format. Painless, without nasty chemistry.
If you need very long fragments, it won't do that, but is good for producing 50 kb pieces. Variations handle tissue, plant, blood.

<*>Thanks veteran, actually i need the gDNA and plasmid for whole genome sequencing, that means i want large good quality gDNA and plasmid.

Typical modern whole genome sequencing involves, for the first step, shearing the DNA into short fragments. It makes no difference what size fragments you start with. In fact, it is easier to handle the shorter fragments, since they can be pipetted.
